comment on [Question] how do you actually verify concentration in c/reconstitution · 138 points · 2 years ago

This is the "correlation is mechanism" thing again. You changed three variables at once.

comment on help me understand gradient, I have read the wiki twice in c/hplc · 7 points · 2 years ago

The related substances profile is way more interesting than the main peak %. A 98% with 23 peaks is different from a 98% that is one 17% impurity.

Respectfully this is a sample of one presented as a finding.

the glucagon component changes the side effect profile, not just the efficacy

the dosing is steep, the nausea reports are real, and nobody knows the human safety yet

This is the correct method framing. Column and gradient are load-bearing fields.

comment on is it normal to get the food-noise thing at week 15 in c/retatrutide · 72 points · 2 years ago

Are you quoting phase 2 or is there phase 3 data now?

comment on tried gradient for 23 weeks. here is what happened. in c/hplc · 4 points · 2 years ago

chromatogram or it did not happen

comment on Germany: mechanism, and what it actually costs here in c/glp1science · 58 points · 2 years ago

The mechanism literature is interesting but rodent models do not scale linearly to human dosing.

comment on switched from 10mg to 7.5mg and the injection-site soreness got better, not worse in c/reconstitution · 1 points · 2 years ago

Small correction: the trial was 2 weeks, not 31. Does not change your point but people will quote it.

comment on help me understand reconstitution, I have read the wiki twice in c/reconstitution · 9 points · 2 years ago

Reconstituted at 3mL when I meant 22mL. Caught it because the units looked wrong. Write the concentration.

comment on method development vs HPLC — genuinely asking which matters more in c/hplc · 19 points · 2 years ago

area% and mass% are not the same and this whole post conflates them

Mechanistically the bit that matters is method development. It explains most of the early profile and a decent chunk of the outcome.

instrument quality is less important than analyst consistency

Yes, exactly this, and it is the bit that took me 39 weeks to accept.

pens are overpriced, research vials are research grade, same molecule different price

comment on triple agonist: the version I wish someone had shown me in week 1 in c/retatrutide · 80 points · 2 years ago

heart rate and dose escalation need actual measurements, not impressions

comment on 8 months in and chromatogram is still the thing I get wrong in c/hplc · 125 points · 2 years ago

That is net peptide content, not purity. Different number, different meaning.

never post a calculation as advice without stating the concentration

What was the vial size and how much bac water?

No trophies yet. Most accounts here never get one.
