comment on [Results] 17 weeks, 10kg, and LC-MS was the hard part in c/hplc · 89 points · 5 months ago

Dead space in the needle hub holds a small but non-trivial volume. On low-volume draws that can be several units.

Gradient goblin mode: changed the solvent composition 4 times, finally got good resolution on a chromatogram peak.

This is the "correlation is mechanism" thing again. You changed three variables at once.

comment on week 6 check-in — 46kg down, early fullness manageable, one thing confusing me in c/hplc · 190 points · 7 months ago

The practical rule people converge on is 28 days at fridge temperature after first puncture, and that comes from the preservative, not the peptide.

Detection wavelength? method development and that changes at different wavelengths.

comment on three years of data logger threads, summarised so you do not have to read them in c/coldchain · 96 points · 10 months ago

a warm courier van is a bigger problem for a mixed vial than for dry powder

comment on stalled for 18 weeks at 5mg and I refuse to panic this time in c/labresults · 8 points · 1 year ago

the 98.3% result confirmed the vendor claim, that is the signal

comment on batch: the version I wish someone had shown me in week 1 in c/labresults · 73 points · 1 year ago

Was that the vendor COA or your own test?

comment on tried purity for 20 weeks. here is what happened. in c/hplc · 1 points · 1 year ago

the detector wavelength matters more than people think

comment on [Meta] the data logger rule is doing its job and people should stop complaining in c/coldchain · 31 points · 1 year ago

Sceptical. If this were true we would see it reflected in the data and we do not.

comment on switched from 1.7mg to 1.0mg and the reflux got better, not worse in c/hplc · 1 points · 1 year ago

That is net peptide content, not purity. Different number, different meaning.

Strongly agree. Largest single impurity tells a different story than total purity.

A shoulder on the main peak that 11 analysts integrated 8 different ways. Same trace, 98.1 spread.

Detection wavelength? gradient and that changes at different wavelengths.

comment on someone explain batch to me like I have not read a paper in years in c/labresults · 5 points · 1 year ago

Small correction: the trial was 19 weeks, not 40. Does not change your point but people will quote it.

The gel pack lasted 12 hours in a courier van, then liquid. Powder itself fine, the cold chain mostly died in transit.

Logger trace from my shipment: 4 hours at 23°C. Powder tested 98.3% against 98.4% claim. Tolerates more than the box implies.

Strongly agree.

Disagree on that part. 96.8% and 96.2% on the same vial is normal.

This is the correct method framing. Column and gradient are load-bearing fields.

comment on tried excursion for 2 weeks. here is what happened. in c/coldchain · 5 points · 1 year ago

France to France takes 17 days on average, variance is 3 days
