Speculationbranch of 5 comments

Flairing this Speculation because that is what it is. Orforglipron is a small molecule, not a peptide. ATTAIN-1 and ACHIEVE-1 (both NEJM , 2025) reported the efficacy numbers everyone quotes, but the interesting property is not the percentage. It is that a non-peptide oral escapes the absorption problem that makes…

The analytical half of this is a bigger change than people assume. Small-molecule purity work has different failure modes — you are looking for process impurities and degradants with well-defined structures rather than truncations and deamidations across a 30-residue chain.

It is also, generally, easier and cheaper to characterise. Which cuts both ways: easier for legitimate suppliers and easier for anyone generating a plausible-looking document.

i had not thought about the forgery side. a simpler assay is a simpler thing to fake convincingly.

and the community has three years of pattern-matching built on peptide documents. we would be starting again on the tells.

the nine patterns are mostly about documents and behaviour rather than chemistry, so i think most of them port. the chemistry-specific ones would not.

fair. the recycled-COA and generated-social-proof patterns are chemistry-agnostic.

About c/orforglipron

The small-molecule oral: why a non-peptide agonist escapes the absorption problems of oral semaglutide, what the ATTAIN and ACHIEVE readouts showed, and what a pill with no refrigeration requirement would do to the entire supply conversation this site is built around.

  1. Small molecule, not a peptide. Purity discussion here is a different problem.
  2. Cite ATTAIN/ACHIEVE arms specifically when quoting numbers.
  3. Availability speculation must be flaired Speculation.
  4. Independent community. Nobody here sells anything, and anyone who tries is banned.
  5. Not medical advice. Describe what you did; never prescribe to a stranger.
  6. Claims need evidence. Batch numbers, dated screenshots, independent test reports, or a citation.
