anyone else notice endpoint kicking in around week 19

Readout Well Actually ×3 Clean Column ×1

Genuinely asking, not being difficult.

Everyone in c/compounding repeats that hazard ratio is important. I believe it, but I have never seen anyone show why, and when I search I get 2 threads of people agreeing with each other.

Is there a paper? Is there a measurement? Or is this one of those things that is true because it has been repeated since 2023?

I am not trying to be the "source?" guy. I would just like a source.

STEP, SURMOUNT, SURPASS, SELECT, FLOW, TRIUMPH — read the appendix

The confident tone is doing a lot of work that the evidence is not.

number-needed-to-treat is the house dialect

Titration speed is a side-effect dial far more than an efficacy dial.

Water content matters more than people think. If a vial is carrying residual moisture, your "10mg" is not 10mg of peptide.

off-label use data is different from approved-indication data

Yes, exactly this, and it is the bit that took me 91 weeks to accept.

Is that absolute or relative risk reduction?

Yeah, absolute numbers matter more than the press release.

Phase 2 or phase 3?

the 7 week readout is different from the 13 week readout

the relative risk reduction in headlines is not the whole story

Adding to this: hazard ratio is doing more work than the comment implies.

event rates are more informative than hazard ratios alone

give the trial name, phase, n, and primary endpoint in the body

STEP 1 readout: 99.3% mean weight loss, absolute numbers matter. Spread around that mean is enormous.

Crossposting this to c/glp1muscle because the people who need it are not reading this community.

Phase 2 TRIUMPH looked amazing. Phase 3 trials are harder. Do not anchor on phase 2 readouts.

the discontinuation arms are the interesting part, not the SURPASS endpoints

press release SURMOUNT posts must be flaired until a publication exists

Disagree but this is the good kind of wrong — it is specific enough to be checked.

This is correct. Discontinuation arms show the regain reality.
