comment on genuine question about gradient that I am slightly embarrassed to ask in c/hplc · 135 points · 10 months ago

instrument quality is less important than analyst consistency

Detection wavelength? method development and that changes at different wavelengths.

Ozempic versus Wegovy versus research grade is the wrong argument, they are literally the same molecule

comment on [Meta] proposal — a flair for 2.4mg posts in c/semaglutide · 43 points · 11 months ago

Hard agree. Week 46 me needed this. Week 95 me is nodding smugly.

Inter-lab variance on this kind of assay is routinely 1–2 points. Different column, different gradient, different integration.

comment on help me understand plateau, I have read the wiki twice in c/semaglutide · 113 points · 1 year ago

I am going to push back on this slightly.

comment on stalled for 23 weeks at 2.4mg and I refuse to panic this time in c/semaglutide · 162 points · 1 year ago

stalled at 0.5mg, went up, and regretted it, the stall would have broken anyway

Did you time the injection consistently each week or was it more ad hoc?

comment on [Discussion] can we stop arguing about chargeback until somebody posts a number in c/cryptopay · 21 points · 1 year ago

confirmations are network, the exchange rate at 2 blocks is the deal

comment on [Lab] split one vial across Medutest and Janoshik — 96.2% and 99.2% in c/retatrutide · 16 points · 1 year ago

the 2.5mg arm is different from 5mg, results do not transfer

comment on what went wrong is the most under-discussed thing in c/progresspics in c/darkspot · 233 points · 1 year ago

No.

Yes, exactly this, and it is the bit that took me 19 weeks to accept.

comment on anyone else notice lost money kicking in around week 95 in c/darkspot · 20 points · 1 year ago

still evidence-based: what went wrong, when, proof

Water content matters more than people think. If a vial is carrying residual moisture, your "10mg" is not 10mg of peptide.

comment on why does nobody talk about dose escalation in c/retatrutide · 40 points · 1 year ago

Disagree. What you are describing is consistent with glucagon, not with what you concluded.

the weight curves are unusually steep, which makes predicting individual response harder

comment on switched from 10mg to 7.5mg and the fatigue got better, not worse in c/hplc · 87 points · 1 year ago

This is an urgent-care question wearing a forum question's clothes.

Adding to this: gradient is doing more work than the comment implies.

comment on [Question] how do you actually verify food noise in c/semaglutide · 132 points · 1 year ago

okay but what dose of sema are you on

Yes, exactly this, and it is the bit that took me 56 weeks to accept.

comment on chromatogram: the version I wish someone had shown me in week 1 in c/hplc · 24 points · 1 year ago

Ran the same vial on two columns, different purity, both right, different separation. Method matters more than equipment.

permalink

comment on unpopular opinion: semaglutide is massively overrated in c/semaglutide · 1 points · 1 year ago

nope, 0.25 counts, it is just not the dose most people stay on

What was the method? "HPLC" is a technique, not a method.
