comment on regulatory is the most under-discussed thing in c/vendorvetting in c/bpc157 · 2 points · 1 year ago

near-zero controlled human data, full stop

Disagree but this is the good kind of wrong — it is specific enough to be checked.

the weight curves are unusually steep, which makes predicting individual response harder

comment on small win: tapering stopped being a problem at week 30 in c/tapering · 49 points · 1 year ago

What are you comparing it against though?

comment on [Discussion] we are measuring T2D at the wrong time and calling it noise in c/t2dglp1 · 11 points · 1 year ago

Inter-lab variance on this kind of assay is routinely 1–2 points. Different column, different gradient, different integration.

say how long you were on and at what dose before taper

comment on the regret thing finally clicked for me and I want to write it down in c/darkspot · 51 points · 1 year ago

Titration speed is a side-effect dial far more than an efficacy dial.

interval stretching versus dose reduction

comment on lyophilised vs stability — genuinely asking which matters more in c/coldchain · 12 points · 1 year ago

Yeah, data logger is the only acceptable proof.

no tracking numbers, no full addresses, no label photos — instant removal

Counter-anecdote: opposite result, same dose. Which mostly tells us the variance is huge.

This is what I tell everyone in c/newbiequestions and they never believe me until week 47.

heart rate and dose escalation need actual measurements, not impressions

glycaemic control first, that is the original indication

Dose escalation on retatrutide is steeper and faster than on semaglutide. Do not assume the titration patterns transfer.

The glucagon component does different things than dual agonism. The the food-noise thing are not just amplified versions.

TRIUMPH phase 2 showed 99.3% weight loss but the the food-noise thing profile on the highest arm was rough. Phase 2 != phase 3.

stalled at 5mg, went up, and regretted it, the stall would have broken anyway

Yeah, rodent data does not scale to humans.
