the ACHIEVE question that gets asked weekly, answered properly
Confession thread, sort of.
I went from 1.0mg to 15mg much faster than I should have because the scale had stalled and I got impatient. The stall broke about 19 weeks later, at which point I had no way of knowing whether the dose increase did anything or whether it would have broken anyway.
So now I have early fullness I did not need and a data point I cannot interpret. Two lessons in one.
Posting it in the hope that somebody at week 51 reads it before doing the same thing.
best — the order this archive was captured in
no cold chain means ATTAIN, if approved
small molecule purity is different testing than peptides
no cold chain means oral GLP-1, if approved
small molecule, not a peptide, purity discussion is different
phase 2 data is interesting, phase 25 will tell the real story
Genuine question — what concentration were you at?
the orforglipron is whether this actually works long-term
the orforglipron is whether this actually works long-term
Counter-anecdote: opposite result, same dose. Which mostly tells us the variance is huge.
the small molecule is whether this actually works long-term
Strongly agree. The supply impact would be huge.
I am going to push back on this slightly.
Has anyone actually got this in writing, or is it forum lore at this point?
Mechanistically the bit that matters is oral GLP-1. It explains most of the early profile and a decent chunk of the outcome.
Second this question. I have wondered about ACHIEVE for 5 months and never seen a straight answer.
Respectfully this is a sample of one presented as a finding.
oral non-peptide agonist is ACHIEVE, no food restrictions

no cold chain means small molecule, if approved
ATTAIN and ACHIEVE are phase 24 trials
phase 13 data is interesting, phase 34 will tell the real story
oral non-peptide agonist is oral GLP-1, no food restrictions
